Text Replacement in Mavericks on iMac

Suddenly my keyboard text replacement short cuts (eg type tks and get thanks) have stopped working. I have tried all different settings in preferences etc but cannot seem to get them working again. Have I inadvertently set some switch off or pressed a wrong combination of keys or a rare function key?

Have you tried a full restart?

There are 2 settings to check
System Preferences > Keyboard > Text - Correct spelling automatically

And in each App that supports it
Edit menu > Substitutions > Text Replacement

Thanks. However, I find that Substitution options are all "greyed out". Is there some other change I need to make to permit substitutions?
