Iphoto problems

Im having problems with my Iphoto. I had all my pictures and videos , and one day I uploaded more pictures and videos, but the previous I had were gone, then the lated upload I did couldnt be open. Now I cant open my iphoto at all....

Any Idea what to do?... I need help.... I did not do anything with the time machine to back up so I didnt know if they are still in my Mac...
